چکیده

The spread of influenza A viruses is partially controlled and prevented by vaccination. The matrix protein 2 ectodomain (M2e) is the most conserved sequence in influenza A viruses, and is therefore a good potential target for a vaccine to protect against multiple virus subtypes. We explored the feasibility of an M2e-based universal influenza A vaccine candidate based on the highly pathogenic avian influenza A virus, H5N1. A synthetic M2e gene was human- and plant-codon optimized and fused in-frame with a sequence encoding the N-terminal proline-rich domain (Zera(®)) of the γ-zein protein of maize. Zera(®)M2e was expressed transiently in Nicotiana benthamiana and Sf21 baculovirus/insect cell expression systems, and Zera(®)M2e protein bodies (PBs) were successfully produced in both expression systems. The plant-produced Zera(®)M2e PBs were purified and injected into Balb/c mice. Western blot analysis using insect cell-produced Zera(®)M2e PBs and multiple tandem M2e sequences (5xM2e) fused with the avian influenza H5N1 transmembrane and cytosolic tail (5xM2e_tHA) confirmed the presence of M2e-specific antibodies in immunized mice sera. The immunogenicity of the Zera(®)M2e indicates that our plant-produced protein has potential as an inexpensive universal influenza A vaccine.

Expression of HA1 antigen of H5N1 influenza virus as a potent candidate for vaccine in bacterial system

The impending influenza virus pandemic requires global vaccination to prevent large-scale mortality and morbidity, but traditional influenza virus vaccine production is too slow for rapid responses. In this study, bacterial system has been developed for expression and purification of properly folded HA1 antigen as a rapid response to emerging pandemic strains. Here, a recombinant H5N1 (A/Indone...

Construction of a Recombinant Bacmid DNA to Express Influenza Virus Matrix Protein1 (M1) in Insect Cell Line

Background and Aims: Virus-like particles (VLPs) have been suggested to be a promising recombinant vaccine approach. Several studies have reported that the influenza VLPs produced in insect cells is an effective vaccine candidate. Due to crucial role of matrix M1 protein in assembly and budding of Influenza particles, in all VLPs structures, M1 protein have been considered as a main component. ...

Construction of chimeric protein 3M2e.FliC and its immunoinformatics analyses and expression in Bacillus subtilis

Introduction: Influenza A virus causes unpredictable epidemics and pandemics by creating antigenic variations. With the appearance of each new strain, rapid emergency countermeasures are taken against this new strain. Hence, designing an applicable and cross protective strategy to counter this virus is of great importance. To achieve this, choosing conserved antigenic regions in influenza virus...

Cloning of the Gene Encoding M2e of Influenza Virus in B. subtilis

Background and Aims: The ectodomain of matrix protein of influenza virus is a weak immunogen that is highly conserved among all subtypes of influenza A virus. Tandem repeats of these genes along with linker were used to enhance immunogenicity of M2e protein and so it can be served as a universal vaccine in both humans and livestock. Materials and Methods: In this study, the sequences of extra-d...

Over Expression of Influenza Virus M2 Protein in Prokaryotic System

Background and Aims: Influenza A virus of Orthomyxoviridae family is able to create pandemic influenza. Vaccination is the most effective way to prevent influenza virus infection. Matrix protein 2 (M2) is a homotetramer ion channel with 97 amino acids length and highly conserved among influenza viruses and is considered for development of a universal influenza vaccine. Materials and Methods: We...
